UDС 615.065:54.061/.062:547.712.22:001.8 ANALYTICAL DIAGNOSTICS OF ACUTE SULPIRIDE POISONINGS S.V.Bayurka, V.V.Bolotov, S.A.Karpushina, V.I.Stepanenko, O.G.Pogosyan

The effective methods of liquid-liquid extraction of sulpiride from the biological fluids with ethyl acetate from the alkaline medium at pH 10-11 have been developed. Identification of sulpiride in the biological extracts obtained was carried out by the Marquis test and Froehde reagent, Thin Layer Chromatography using such mobile phases as methanol – 25% solution of ammonium hydroxide (100:1.5) and benzene-methanol-diethylamine (90:10:10). The Rf values were 0.53±0.02 (for HPTLC plates), 0.55±0.02 (for Sorbfil plates) and 0.60±0.02 (for HPTLC plates), 0.52±0.02 (for Sorbfil plates), respectively. After elution of sulpiride from the plates with methanol UV-spectroscopy detection of the medicine was carried out (λmax 293±2 nm). The quantitative determination of sulpiride in the extracts was performed by UV-spectrophotometry. The methods developed allowed to isolate 64.6±3.5% of sulpiride from the urine, 16.9±2.2% from the blood serum and additionally 7.7±1.1% of the antidepressant studied from the blood sediment after its separation from the blood serum.
